Zofran, also marketed as ondansetron by GlaxoSmithKline, is a drug used to treat nausea. It is commonly given to individuals undergoing chemotherapy for cancer. Zofran blocks serotonin, a chemical that acts on the nerves in the gut to produce feelings of pain and nausea. It also increases the speed of food leaving the gut to reduce nausea and vomiting. Previous research has established that serotonin, along with another neurotransmitter, stimulate the reorientation of the palate in the embryo. It is likely that taking a drug like Zofran inhibits serotonin’s ability to correctly reorient the palate in the embryo, leading to mouth deformities like cleft palate.

Cleft palate is very disfiguring and usually must be corrected surgically, either in utero or after birth.
